1914年7月一战前夕，东方列车踏上了从巴黎开往君士坦丁堡的最后一趟旅程，玩家化身为美国医生罗伯特·凯斯，应挚友泰勒·惠特尼之邀，登上东方列车与其会合。为了弄清楚为什么泰勒严词要求自己登上东方列车，罗伯特·凯斯发现自己的挚友死在车厢里，他身上携带着一幅神秘的俄文卷轴。从隔壁车厢到餐厅，对火车上的所有车厢一一进行调查。
